L\u2019utilisation accrue d\u2019antibiotiques a favoris\u00e9 l\u2019\u00e9mergence de souches de K. pneumoniae r\u00e9sistantes aux carbap\u00e9n\u00e8mes et productrices de \u03b2-lactamases (ESBL). R\u00e9cemment, la phagoth\u00e9rapie a gagn\u00e9 en dynamisme en tant qu\u2019alternative potentielle face \u00e0 la r\u00e9sistance antimicrobienne \u00e9mergente. Cette \u00e9tude a \u00e9t\u00e9 men\u00e9e afin d\u2019\u00e9valuer les effets th\u00e9rapeutiques d\u2019un nouveau phage lytique (VTCCBPA43) dans un mod\u00e8le murin de pneumonie, et d\u2019examiner l\u2019efficacit\u00e9 de la phagoth\u00e9rapie contre une infection virulente \u00e0 K. pneumoniae.   <\/p>\n<p>M\u00e9thodes<br \/>\nLe phage \u00e0 queue VTCCBPA43 a \u00e9t\u00e9 \u00e9tudi\u00e9 pour sa cin\u00e9tique de croissance, son spectre d\u2019h\u00f4tes in vitro, ainsi que sa sensibilit\u00e9 \u00e0 la temp\u00e9rature et au pH. Les composants prot\u00e9iques ont \u00e9t\u00e9 analys\u00e9s par SDS-PAGE et nLC-MS\/MS. L\u2019efficacit\u00e9 th\u00e9rapeutique a \u00e9t\u00e9 observ\u00e9e 2 h apr\u00e8s l\u2019\u00e9preuve avec une souche virulente de K. pneumoniae dans un mod\u00e8le murin BALB\/c.  <\/p>\n<p>R\u00e9sultats<br \/>\nLe phage VTCCBPA43 s\u2019est r\u00e9v\u00e9l\u00e9 tr\u00e8s tol\u00e9rant \u00e0 la temp\u00e9rature (jusqu\u2019\u00e0 80 \u00b0C). Il \u00e9tait le plus actif \u00e0 pH 5, pr\u00e9sentait une taille de burst de 172 PFU\/mL et un spectre d\u2019h\u00f4tes \u00e9troit. Il a \u00e9t\u00e9 identifi\u00e9 par prot\u00e9omique shotgun comme un phage de type KP36. Apr\u00e8s administration intranasale d\u2019une dose unique (2 \u00d7 10^9 PFU\/souris) apr\u00e8s infection par une souche virulente de K. pneumoniae, la pr\u00e9sence d\u2019un phage biologiquement actif in vivo ainsi qu\u2019une diminution significative de la charge bact\u00e9rienne pulmonaire \u00e0 tous les temps ont \u00e9t\u00e9 observ\u00e9es.
